癌における転写制御変異の統合的解析

Research Abstract

1)染色体機能解析
肝細胞癌100症例のメチル化プロファイルでは4群に分かれ、メチル化情報に基づく層別化の可能性が示された。癌で異常メチル化を生じる領域はES細胞でPolycomb複合体が結合し、bivalentなドメインを形成する場合が多く、癌の由来臓器により標的遺伝子が異なることが明らかとなった。
幹細胞から成人細胞への分化時のDNAメチル化の変動について、16種類のヒト成人及び胎児組織のDNAについて約27,000箇所のCpGサイトを調べたところ、GC含量の低いプロモーター領域は非CpGのメチル化も含めてES細胞では高メチル化状態にあり、iPS細胞では再び高メチル化レベルにリプログラムされていた。低CpGプロモーターは組織特異的な発現を示す遺伝子に多く、分化に従って低メチル化がプロモーター領域から次第に拡大することが認められた。
ChIA-PET法によるクロマチン相互作用の同定、chromatin accessibilityを反映するFAIRE解析(Formaldehyde Assisted Isolation of Regulatory Elements)を立ち上げてデータ収集した。
2)転写制御ネットワークの統合解析
肝細胞癌38例についてexon capture法によって約1200遺伝子のエクソン領域のみを約300Xで解析した。β-catenin、TP53はそれぞれ11例、10例と比較的高頻度に変異が認められたが、ほぼ相互排他的に認められ、癌細胞が依存するシグナルの活性化や不活性化に関わる遺伝子変異は症例毎に異なっていた。
膵臓癌15例の全exomeによる変異解析ではKRAS(100%)、SMAD4(60%)、TP53(80%)、P16(67%)の4遺伝子の変異が高頻度に認められ、変異プロファイルという見地からは膵癌の遺伝的成因は比較的均一であると考えられた。
