Research Abstract

核内受容体HNF4αについて高親和性の抗体磁気ビーズによる免疫精製および高感度ショットガンMS解析法を開発し、内在性のHNF4α複合体を培養ディッシュ1枚から定量的に同定した。HNF4αのアイソフォームやりん酸化等の修飾を見出し、またHNFγとヘテロ2両体を形成して糖代謝に重要なCIDE-B遺伝子等を制御していることを明らかにした。さらに、相互作用するクロマチンリモデリング複合体やHATコンプレックスを同定し、新規の転写コファクター候補を同定した。mRNAの安定性やオルタナティブスプライシングに関係するWTAPについて,相互作用するタンパク質を同様にHUVEC細胞から同定し、4種類のタンパク質について抗体を作製し,細胞周期や核内局在に関する解析を行った。これらの複合体構成タンパク質がWTAPのスペックル局在に関与することを明らかにした。また,標的RNAの網羅的解析のため,Par-Clip法を開発した。同定されたRNA、特にnon-coding RNAについて解析を行った。核内受容体LXRαについて、特異抗体を用いた蛍光染色により、HepG2細胞において、核小体への分布があることを突き止めた。リボゾーマルRNAの転写調節との関連が示唆された。
軟X線顕微鏡による観察法を開発するため,核膜タンパク質エメリンに対する抗体を作製した。また、軟X線顕微鏡に適したシリコンナイトライドを用いたサンプルホルダーを開発して細胞を固定し、エメリン抗体および金属コロイド付加2次抗体を用いて、Hela細胞の簡易型軟X線顕微鏡による撮像に成功した。
