多核金属触媒の創製を基盤とする医薬合成の革新

Research Abstract

多核希土類-糖リガンド錯体が触媒的不斉プロトン化反応に極めて有効であることを見出した。本反応は、世界的な脅威となっている多剤耐性結核菌に対して有効なR207910の触媒的不斉全合成へ応用可能で、現在本プロトン化反応を鍵工程とする効率的合成ルートの確立を目指し鋭意検討中である。
2つの金属イオンを取り込むことが可能なシッフ塩基配位子のさらなる展開として、新規Ga/Yb型2核金属触媒の創製に成功した。本触媒はイソシアニドのアルデヒドへの触媒的不斉反応に著効を示し、新たな異種2核金属触媒の可能性を示すことができた。また本シッフ塩基配位子系において同種核Ni、Mn錯体がβ-ケトエステルの不斉アルドール反応やオキシインドールのニトロオレフィンへの不斉付加反応に極めて有効であることが判明し、またこれらの反応で得られる生成物は光学活性医薬品の有用なキラルビルディングブロックとして有用であることから、合成化学的価値が極めて高いと言える。予測手法の精度を高めていく。
アミド型配位子触媒を用いる触媒的不斉ニトロアルドール反応の超高効率化に成功した。論理的にデザインされたアミド配位子とNd/Na異種2核金属触媒を用いることにより、不均一系触媒が自己組織化により形成され、その各種分光学的解析により、両金属を含有する集積型不均一触媒創製がなされている事がわかった。本反応により光学活性1,2-アミノアルコールの効率的触媒的不斉合成が可能になり、現在PhaseIIIにあるβ_3アドレナリン受容体選択的アゴニストの工業的合成への応用研究が進行中である。
