Research Abstract

ヌクレオチド除去修復(NER)の開始段階に必須であるDNA損傷認識因子XPCについて、我々が既に見出した翻訳後修飾(ユビキチン化、SUMO化)に着目し、それぞれの修飾を受けるリジン残基の特定、および修飾・脱修飾に関わる因子の同定を行う。
決定された修飾部位に変異を導入したXPCの組換えタンパク質をバキュロウイルス系を用いて発現・精製し、生化学的な性状変化を解析するほか、これらの変異体を安定に発現する形質転換細胞株を単離して、細胞レベルでのNER活性、DNA損傷に伴うタンパク質の安定性や動態変化、およびチェックポイントや細胞死誘導に関わるシグナリングに対する変異の影響などを体系的に調べる。また、修飾部位を特異的に認識する抗体の作成やGFP再構成システムを構築することにより、それぞれの修飾の時空間的動態制御を観察するとともに、相互の機能的連関を明らかにする。さらに、修飾タンパク質と特異的に相互作用する因子を探索することにより、修飾が直接影響を与える細胞機能について分子レベルでの知見を得る。
