Research Abstract

我々は、マウス大脳皮質の興奮性グルタミン酸作動性錐体(Py)神経細胞と抑制性GABA作動性Parvalbumin(Pv)神経細胞で発現している転写産物を、次世代シークエンサーであるイルミナ社ゲノムアナライザーを用いて、網羅的に解析した。この際、脳の可塑性を再活性化することがわかっている(未発表)、バルプロ酸(VPA)とtricostatin A(TSA)による処理も合わせて行い、トランスクリプトームの変化を調べた。今年度行った、神経細胞調製とRNA抽出についてのプロトコール改変(投稿準備中)と96穴プレートを用いた並列nanoCAGEプロトコルの標準化(Salimullah et al. 2011;doi:10.1101/pdb.prot5559)のおかげで、現在までに、Py神経からは195,459,905個の、Pv神経からは275,065,042個のシークエンスタグを得ることができた。ここから、それぞれの神経細胞で発現する、900万ヶ所以上(Py)と2600万ヶ所以上(Pv)の転写開始点を同定した。これらのデータは、これまでに単離されたいかなる神経細胞においても実現され得なかった、最も網羅的なトランスクリプトームデータを提供する。このデータを使って、Py神経細胞における可塑性の再活性化に伴って発現量の変化する遺伝子の予備的な候補を絞り込み、そのオントロジー解析の結果と合わせて2010年度日本分子生物学会年会にて発表した(Noro et al.poster#:2P-1186)。今後は、今年度投稿、受理されたCAGEscan技術(Plessy et al. Nature Method, 2010;7(7):528-34)を用いて、シークエンスタグの3'側も読み、転写開始点や発現量だけではなく、転写産物の構造をも明らかにできると考えている。すでに、両側読みされたシークエンスタグは、Py神経から60,270,789個、Pv神経から53,439,327個得られており、これらの網羅的データから、可塑性の再活性化に伴って変化するRNAプロセシングの全容を解明していきたい。尚、今年度は、国内6件、海外5件の口頭・ポスター発表により、本研究の成果が所外に公開された。
