アセチルグルコサミン糖鎖サイクルの生体制御機構の解析

Research Abstract

本研究は「糖鎖サイクル」という糖鎖の一生を統合的かつ動的にとらえる概念を提案し、疾患との関わりについて明らかにすることを目的とする。
最終年度は、昨年度までに検討した複合糖質の質量同位体分布解析法により、糖ヌクレオチドがどの糖鎖にどの程度取り込まれたか、その全貌を追跡した。糖鎖の合成過程が少ないハイマンノース型N型糖鎖は複合型糖鎖に比べて高いラベル効率を示したことから、これらの結果は糖鎖の合成速度を反映することがわかった。合成過程がシンプルなヒアルロン酸、O型糖鎖は合成速度が速かった。N型糖鎖の分岐の程度、シアル酸の結合様式の違いにおいて差は認められず、予想に反して、コアフコースを有するN型糖鎖は合成速度が速いこともわかった。
神経芽細胞腫細胞株からGlcNAc転移酵素(GnT-IX)の酵素活性調節因子(エクトヌクレオチドピロホスファターゼ/ホスホジエステラーゼ3: ENPP3)を同定した。その酵素学的な解析から、ENPP3によりUDP-GlcNAcが加水分解されて生じるUMPが、UDP-GlcNAcに対する競争的阻害因子として働いてGnT-IXの活性を阻害していることを明らかにした。
GnT-IIIとコアフコース転移酵素(FuT-8)において補完的な機構があることを突き止めた。具体的にはFut-8欠損マウスの繊維芽細胞や血清中の免疫グロブリンの糖鎖を定量解析した結果、GnT-IIIにより作られるバイセクティングGlcNAcを有する糖鎖が3倍に増加していた。野生型に比べてGnT-IIIの発現に関わるmRNAが3倍に、その酵素活性も8倍に増加していた。原因を解明するため、Fut8欠損MEF細胞の遺伝子変化を調べたところ、同細胞内でシグナル伝達物質であるWntのターゲット遺伝子群が増加していること、また遺伝子の転写を活性化するβカテニンが蓄積していることがわかった。
