インフルエンザウイルスゲノム機能の制御に関わる宿主因子の同定と機能解析

Research Abstract

本研究の目的は、インフルエンザウイルスのゲノム複製と転写に関わる宿主因子を同定し、それらの機能を明らかにすることである。
上記の目的のために、我々はインフルエンザウイルスゲノムの複製と転写を再現できる試験管内系を確立し、その解体と再構成により、宿主因子の探索と機能解析を進め、数種のウイルスRNA合成促進宿主因子を同定した。これまでに、プライマー非依存的な複製反応に関わる宿主因子IREF(Innuenza virus REplication Factor)-1の機能解析をすすめ、IREF-1はminichromosome maintenance(MCM)であり、MCM存在下でウイルスポリメラーゼはProcessiveなRNA合成を行ない、全長のウイルスゲノムを複製することが明らかになった。複製反応に関わる第2の宿主因子としてIREF-2を同定した。IREF-2は、複製中間体であるcRNAを鋳型としたウイルスゲノム複製を特異的に促進する宿主因子であり、鋳型極性を認識する活性をもつ因子であった。本年度では、ウイルスゲノムに結合し、Ribonucleoprotein(RNP)複合体の形成に必須であるウイルスタンパク質NPによっても、ウイルスゲノム複製は促進されることを見出し、NPはウイルスポリメラーゼが開始反応から伸長反応へ移行するのに必要であることを明らかにした。さらに、この過程にはNPの分子シャペロンとして機能する宿主由来のスプライシング関連因子であるRAF-2p48/UAP56が促進因子として機能し、子孫ウイルスRNP複合体形成と協調してRNA合成反応を促進することを明らかにした。また、複製後のウイルスゲノムは、細胞核内から細胞質へと輸送され、細胞膜の頂端面から出芽するが、その詳細な輸送機構は不明であった。感染細胞内のウイルスゲノムをウイルスRNPに対する特異的抗体もしくはFISH法で可視化し、その動態解析を行ったところ、ウイルスRNPの細胞内輸送は微小管依存的でありRab11陽性の小胞輸送を介した経路によって能動輸送されていることを明らかにした。
