光電変換機能をもつ超分子複合体の階層的組織化と機能解析

Research Abstract

昨年に続いて、下記1)および2)で示したように、光合成膜構造を模した階層的ナノ組織体を巧みに構築し、基板上で高効率な光電変換機能をもつタンパク質色素複合体のナノデバイスの開発を行った。また、その色素複合体のAFM観察ならびにX線の構造解析の結果と合わせて、光合成の色素複合体の動的構造と機能との相関について明らかにした。
1) アンテナ系タンパク質色素複合体の電極上での組織化と機能解析
アンテナ系タンパク質として紅色光合成細菌、R.ruburmおよびR.Sphaeroidesを培養し、アンテナ色素、クロロフィルならびにカロテノイド色素と構成するタンパク質(LHα&LHβ)を単離精製して、そのアンテナ系タンパク質/色素複合体の再構成を脂質二分子膜(リポソーム膜)中および基板上で行った。そして、この複合体の形成におよぼすタンパク質の極性アミノ酸組成および脂質の組成の役割について、それぞれ、検討を行った。
2) アンテナ系モデルタンパク質を用いた色素誘導体の基板上での組織化とその機能解析
人工的に調製したアンテナ系モデルタンパク質色素複合体の自己組織化を基板(ITOあるいは金電極)上で行った。その確認は、UV-VisおよびCDなどの分光学的手法ならびにAFMを用いた検討から行った。また、それらの複合体の蛍光挙動ならびに基板上での電流応答の比較検討から、クロロフィル色素誘導体間のエネルギーおよび電子移動について検討を行った。
