耐熱化過程におけるゲノムネットワークの解析

Research Abstract

温度変化の分岐点および45℃長期適応時の耐熱進化大腸菌に関して、オリジナル大腸菌からの全ての変異部位について、塩基配列を確定・確認した。
1. タイリングアレイの変異解析結果を生データレベルで統計的再解析し、サンガー法も用いて全変異に対してシーケンスを行って、ゲノム変異を確定した。1塩基置換以外に、挿入と欠損配列も同定した。
2. 突然変か異率上昇に関しては、ゲノム変異解析だけではなく、遺伝学的に薬耐性のプレート実験により突然変異率が2桁上昇したことを検証した。そのため、45度に耐熱進化した大腸菌株がmutatorであることが証明された。
3. 耐熱進化過程では、37度から43度までの進化過程では、Ka/Ks値(分子進化の指標)が1より大きいため、正の選択がかかっていることに対し、45度での適応進化の過程では、Ka/Ks値がほぼ1であるため、中立的であることが分かった。これは、初めての実験的な証拠であると考えられる。
4. 中立的進化過程にもかかわらず、適応度の指標である増殖速度が持続的に増加していることが初めての発見である。これらの結果から、中立進化と適応進化の渡し橋が見つかることが期待される。
大腸菌の耐熱化を更に進め、0.2℃刻みでの耐熱進化で46.6℃、1℃刻みの耐熱進化で46.8℃までの耐熱進化に成功した。44.8℃からの1℃上昇への適応に要する日数は0.2℃刻みでも1.0℃刻みでもほぼ同じであることが確認された。
高温への適応過程で見られる大腸菌の変化についてFCM解析を行い、高温適応初期には細胞サイズ、内部複雑性の大きな細胞が出現し適応と共に細胞ザイズは大きいが細胞サイズあたりの複雑性が小さくなることが確認された。
プロテオーム解析に関しては、その比較対象株を検討し、解析の準備に着手した。
