Research Abstract

本研究は新たに発見した芳香族炭化水素受容体(AhR)による大腸がん抑制機能について、従来のAPC依存的な分解系とは異なるナチュラルリガンド依存的AhR E3ユビキチンリガーゼ活性によるβ-カテニン分解によることを提唱し、これを確立するものである。具体的には以下を検討する。
(1)AhRとAPCの関連性の解析
(2)ASC・AhR二重遺伝子変異マウスによる発がんと炎症との関係
(3)なぜAhR遺伝子欠損マウスは盲腸に発がんするか?
(4)AhRのヒトの大腸がん(盲腸がん)における役割
それぞれの具体的な研究計画に沿って二重遺伝子欠損マウス及び無菌マウスによる個体レベルでの研究、大腸がん培養細胞レベルでの研究、及びヒト大腸がん臨床検体を用いての研究を計画し、総合的な観点よりAhR E3ユビキチンリガーゼと腸内細菌・炎症との相互作用による発がんとその抑制についての研究を進展させる。
