抗結核ワクチンの新戦略:潜伏感染、再燃を制御するワクチンの研究

Research Abstract

結核に対するワクチンとしては、生菌ワクチンであるBCGが現在も用いられているが、その成人肺結核に対する防御効果は否定的であり、潜伏感染患者を始めとする成人用新規ワクチンの開発が急務とされている。我々は成人結核のほとんどが内因性再燃に起因することに着目し、結核菌の再燃を抑制する新規ワクチンの開発を目的として、その候補抗原を検索中である。潜伏期の結核菌が発現するDosR regulon蛋白(48種)および結核菌の再活性化に関与するResuscitation promoting factor(Rpf)蛋白(5種)を候補抗原として、DNAワクチン・ライブラリーを作製した。現在、これらの候補抗原に対するT細胞応答と抗体産生について、2系統のマウスを用いて検討中である。32種類についてT細胞応答のスクリーニングを終えた時点で、C57BL/6で9種類、BALB/cで12種類の抗原がT細胞応答を誘導し、そのうち7種類が両系統でT細胞応答を誘導することができた。加えて一部の抗原について抗体産生を検討したところ、少なくともC57BL/6で2種類、BALB/cで11種類の抗原が抗体産生を誘導できた(2種類が両系統で抗体産生を誘導)。また、同一系統で両方の免疫応答を誘導できた抗原はC57BL/6で1種類、BALB/cで7種類であった。その中でも、Rv3132(dosS/desS)は両系統のマウスでT細胞応答と抗体産生を共に誘導でき、有望な候補抗原であると考えられた。
