Research Abstract |
Malignant gliomas show dismal prognosis and glioma stem cell may play a pivotal role on this issue. We established glioma stem cell lines from surgical samples using spheroid method. Using these glioma stem cells, we biologically analyzed 2 novel molecules, N-myc downstream-related gene (NDRG) and EVI1. (1) Glioma stem cell did not express NDRG. However, after differentiation in response to serum, glioma cell strongly express NDRG. (2) EVI1 is a transcriptional factor. Knock-out mice show abnormality in central nervous system. We have shown that knock-down of EVI1 in glioma stem cell and found that Notch signal transduction system was strongly supressed. These novel findings might be important for establishing new treatment against malignant glioma.
