Research Abstract

網膜疾患時の組織破壊物が積極的に生物学的作用をおよぼすことを目的とした研究を行い、昨年蛋白デグラドームのひとつであるHMGB1が網膜環境維持に重要であると報告した(Arimura,Sakamoto et al. Lab Invest 2009, Ophthalmology 2009など)。その結果、マクロファージやSDF-1(stromal cell derived factor)が通説と異なり組織保護に重要であることがわかったので、その点に絞って研究した。人の網膜剥離眼では、網膜剥離範囲、剥離期間に比例して硝子体内SDF-1が増加した。神経網膜細胞R28を用いた研究においては、SDF-1はアポトーシスを抑制し、wound healing assayで細胞保護的に働いた。そこで、in vivo retinal detachment ratモデルを用いてSDF-1の作用を調べたところ、アポトーシス、網膜細胞減少を抑制し、細胞保護効果を示した。この報告はAm J Patholに直ちに掲載されたが、その11月号でSDF-1の組織修復における新たな役割を示す重要研究であるとcommentaryで紹介された(Penn, Am J Pathol 2011)。なおそのほかに、組織保護治療として、Non-steroidal anti-inflammatory drugを用いると、抗酸化蛋白HO-1が増加して、治療効果を持つことを証明して、現在Laboratory Investigation誌に投稿中である。
