放射線の変異誘導を調節するヒトの生理機構におけるシャペロンとマイクロRNAの役割

Research Abstract

本研究は、ヒトにおける異変誘導調節に関する分子生理メカニズムの解明を目的としている。その生理機構は、(1)サイトカイン様血清因子の血液中でのクロストーク的変動→(2)放射線照射直後のプロテアーゼの活性化を伴うシャペロン分子などの発現誘導→(3)核酸代謝の変動→変異誘導の調節である。3年間の研究では(1)、(2)、(3)の分子機構の解明を中心に進め、次の研究項目を実施し、新たな知見を得た。(1)シャペロン結合タンパク・プロテアーゼの解析;HSP27との結合を見出したアネキシンIIが、HSP27と共に細胞の核内へ移入することより、変異誘導を調節するとの示唆を得た(Photochem.Photobio1.,84,1455-81,2008)。(2)変異誘導調節血清因子の同定;シャペロン・プロテアーゼの応答システムを誘導する血清因子の存在が、すでに変異誘導の抑制を見出してあるGRP78を介していることを膵癌患者で見出した(Pancreas,36,e7-14,2008)。(3)新規タンパクの役割とDNA修復連結メカニズムの解明;シャペロン以外のたんぱく類で新規に放射線耐性化に関わるものを見出し、aldolaseA(Biochem.Biophys.Res.Commun.,369,948-52)やglyceraldehyde-3-phosphate dehydrogenase(Biosci.Biotechnol.Biochem.,72,2432-5,2008)と同定した。以上の研究成果は、未だ分子機構が判然としないclastogenic factorやバイスタンダードの効果をも包容した変異誘導調節というヒトの新しいホメオスターシスのメカニズムの一端を解明し、将来、そのホメオスターシスの調節方法が開発され、放射線被爆対処法や制癌などの研究で新たな手法を提供することも期待される。
