放射光真空紫外円二色性による生体分子の高次元構造解析

Research Abstract

広島大学放射光科学研究センターの真空紫外円二色性(VUVCD)分散計を用い、以下の4点について研究した。[1]ジスルフィド結合欠損蛋白質の網羅的二次構造解析:4本のジスルフィド結合を順次欠損した13種のリゾチーム変異体のVUVCDスペクトルを170nmまで測定し、VUVCD-NN法を用いてα-ヘリックスとβ-ストランドの保存状態を解析し、立体構造保持における各ジスルフィド結合の役割を明らかにするとともに、アミロイド形成に関わるβ-ストランドの部位を予測した。[2]多糖類(グリコサミノグリカン)のVUVCDスペクトル解析:コンドロイチン、コンドロイチン硫酸(A, B, C)、ヒアルロン酸、ヘパリンおよびそれらの構成糖のVUVCDスペクトルを160nmまで測定し、COOH, SO_3H, OH, CH_2OH, NHCOCH_3基などの寄与を解析した。[3]酸性糖タンパク質α_1-acid glycoprotein (AGP)の二次構造:AGPはリン脂質との相互作用により、α-ヘリックスは14%(3本)から50%(8本)に、β-ストランドは38%(8本)から5.6%(2本)へと変化し、大きな二次構造変化が誘起されることがわかった。これらの構造変化は主としてリガンド結合部位で起こっており、蛋白質-生体膜相互作用による薬剤の細胞内輸送の機構について重要な知見が得られた。[4]β_2-microglobulinのアミロイドフィブリルの構造解析:赤外・ラマンスペクトルとVUVCDスペクトルを併用することにより、アミロイドフィブリルのlong straight型とworm-like型の2つの構造の違いが,カルボキシル基の解離状態に起因することを明らかにした。
これらの結果は、VUVCD分光法が生体分子の構造解析に有効であることを示している。
