新規気管支喘息治療薬開発に向けた気管支平滑筋過敏性発現メカニズムの解明

Research Abstract

1.気管支平滑筋過敏性発症に関与するサイトカイン受容体の同定 気管支喘息時の気管支平滑筋細胞においてIL13Rα1受容体がup-regulationしており、これによりIL-13 signalingが大幅に増強されており、その結果RhoA transcriptionが亢進してRhoAタンパク質発現が増大することを証明した。
2.気管支平滑筋過敏性発症に関与するSTATサブタイプの同定 マウスアレルギー性気管支喘息モデルに選択的STAT6阻害薬AS1517499をin vivo前処置することにより、気道過敏性およRhoAup-regulationが抑制されることを証明した。したがって、気管支喘息時に気道局所において増加するIL-13が気管支平滑筋に作用し、STAT6依存的にRhoA up-regulationを引き起こし、気管支平滑筋過敏性を惹起する可能性が示唆された。さらに、本モデルマウスの気管支平滑筋においてmiR-133aがdown-regulationしており、これがRhoA up-regulationに一部関与している可能性を示唆した。
3.気管支平滑筋におけるRhoA転写調節領域および転写制御因子の同定 ヒト、マウスおよびラットにおいてRh。A遺伝子のプロモーター解析を行ったところ、転写開始点に最も近いSTAT6 binding領域およびNF-kB binding領域がIL-13誘発RhoA転写活性亢進に関与していることを証明した。さらに、気管支平滑筋細胞においてIL-13刺激によりSTAT6およびNF-kBが実際に活性化されることを証明した。
