Research Abstract

腸炎ビブリオは日本では細菌性食中毒の主要原因菌の一つであり、病原性を発揮するために優れた鉄獲得機構を持つ。申請者らは、腸炎ビブリオが鉄と特異的に結合するキレーターであるシデロフォア・ビブリオフェリンを体外に分泌し、鉄・ビブリオフェリン複合体をPvuAという受容体蛋白を介して取り込む系があることを見いだし、鉄制限に応じた発現制御が行われていることについて明らかにしてきた。今回、申請者は、PvuAの機能を蛋白質化学的に明らかにし、病原性への関与の証明、鉄取り込み阻害剤の開発を目的に、次の検討を行う。(1)PvuAの変異蛋白質を作製し、鉄の取り込みに必要な部位を決定する、(2)PvuAの大量発現株を作製することによりPvuAを大量発現させた後、結晶化し、そのX線結晶構造解析を行う、(3)鉄取り込みを阻害する抗PvuAモノクローナル抗体の調製およびエピトープの解析を行う。
