Research Abstract

非アルコール性脂肪性肝炎(Non alcoholicsteatohepatitis, NASH)の病態における核内レセプターPXRの意義、並びにレセプターCARとのクロストークの詳細を明らかにする。我々は、平成18-19年度科学研究費補助金を得て、CA(constitutive androstanereceptor)のNASHの病態への関与を解明し報告した(Gut.56;565-74:2007)。CARは、肝臓に発現し、薬物代謝酵素(CYP、UGT、MRPなど)の発現調節を行う核内レセプターであるが、糖代謝、脂質代謝にも深く関与している。一方、PXR(pregnane X receptor)も、CARと同様にRXRとヘテロダイマーを形成し、薬物代謝酵素や脂質代謝を制御する核内受容体である。多くの遺伝子プロモーター領域で、CARと結合部位を共有しており、CARとPXRは互いに密接に関連している。メチオニンーコリン欠乏食(MCD食)におけるNASHモデルで、CARの活性化がNASHの増悪因子であり、MCD食の投与で、CARは細胞質から核内へ移動し、標的遺伝子を誘導することによりROS(Reactive Oxygen Species)を産生しNASHを増悪させる。本研究では、PXR及びCARKOマウス、double KOマウスを用い、NASHの病態における核内受容体の関与、PPARも含めた核内受容体相互間のクロストークを解明する。本研究は、NASHの病態の解明だけでなく、CAR及びPXRの生物学的意義の解明であり、臨床医学上非常に意義のある研究と考える。
