C型肝炎ウイルス感染におけるミトコンドリア機能異常の機序の解明

Research Abstract

我々が作成したHCVコア遺伝子トランスジェニックマウスは肝臓脂肪化、肝細胞癌(Moriya et al.Nat Med 1998)、インスリン抵抗性(Gastroenterology 2004)脂質代謝異常(Moriya et al.Biophys Biochem Res Commun 2001)肝臓内酸化ストレス増加(Moriya et al.Cancer Ees 2001)が認められ臨床像に極めて近い。またこれらの病態がミトコンドリア機能異常に起因することを現在まで示してきや。今回コア蛋白によるミトコンドリア機能異常の同定と分子レベルの機序の検討をミトコンドリアプロテオーム解析を用いてを行った。
培養細胞、レプリコン系、トランスジェニックマウスにおいてコア蛋白存在下でミトコンドリアに局在するprohibitinの著名な発現亢進を認めた。Prohibitinはミトコンドリア電子伝達系cytochrome c oxidase(COX)subunitと結合、発現亢進により電子伝達系IV機能低下を示した(Hepatology. 2009)。またミトコンドリア保護機能を有するTacrolimusをトランスジェニックマウスマウス系に投与しコア蛋白による脂肪肝、インスリン抵抗性の軽減を認めた。コア蛋白発現培養細胞系の検討からコア蛋白によるTNFα亢進をTacrolimusが軽減し、ひいては脂質代謝の改善、酸化ストレスの減少をもたらすという新しい治験と、新しい治療法の可能性を示した(Moriya et al.Am J Pathol 2009)。
