IgA腎症患者の扁桃における遺伝子発現

Research Abstract

本研究では、IgA腎症の原因が扁桃腺で産生されるIgA1に関連すると仮説を立て、まずIgA腎症患者群の扁桃と、慢性扁桃炎の患者群の扁桃の遺伝子発現の差異をマイクロアレイ法で検討することでIgA腎症発症にかかわる扁桃の遺伝子発現の特徴を明らかすることを目的とする。研究の方法としては、IgA腎症患者の扁桃と、コントロール(慢性扁桃炎など)の扁桃のRNAをプールし、マイクロアレイ法を用いて比較検討する。この両者で、差があった遺伝子について実際に各症例にもどってその差が有意であるかどうかを検証する。さらに、その遺伝子群の中でIgA腎症との関連が示唆されるものについて、扁桃での遺伝子発現等を検討する。IgA腎症の患者において扁桃摘出を行った扁桃と、慢性扁桃炎において扁桃摘出を行った扁桃で遺伝子発現の違いをマイクロアレイ法により検討し、IgA腎症の患者において約2倍以上の上昇が認められた遺伝子発現として12種あり、筋肉由来以外のものを抽出すると以下のものがあった。2倍以上変化のあった遺伝子の、変化量(Fold Change)と遺伝子名を列挙すると3.44倍:FKSG22,3.28倍:balbindin2、2.12倍:APOBEC2、2.11倍:CEXL11、2.08倍:nebulin、2.07倍:sarolipin、2.08倍fibronectin、1.97倍:Osteopontinであった。このうちAPOBEC2について、遺伝子発現が、IgA腎症患者で実際に有意に上昇し、扁桃において、胚中心の周囲に発現していることを確認した。
