| Research Abstract |
甲状腺ホルモン受容体(TR)、ペルオキシソーム増殖剤活性化受容体(PPAR)、レチノイドX受容体(RXR)は、全て核内ステロイドホルモン受容体ファミリーのメンバーである。TR及びPPARは、各々RXRと2量体を形成して、標的遺伝子の上流にある特定の部位に結合(共通のDNA配列AGGTCAを認識)し、当該リガンドに応答して各々の標的遺伝子の転写を活性化する。各受容体には、複数のアイソフォームが存在するが、その明確な役割分担は不明である。各受容体、各アイソフォームは、お互いに相互作用しうることから、その組み合わせは膨大となり、標的遺伝子毎に極めて多様な作用を発揮していることが想定される。本研究では,これら受容体間、アイソフォーム間の複雑なクロストークを、我々が最近同定した新規TRアイソフォームも含めて、主としてエネルギー代謝と骨代謝の観点から解明することが目的である。さらに、共通のプラットフォームを用い、受容体アイソフォームをベイトとして、生薬の中からアイソフォーム特異的リガンドを新規に同定する。
