Research Abstract

当該年度において申請者は、当初の予定に添って、RT-PCR法を用いてヒト造血幹細胞(CD34+細胞)と系特異的分化誘導を行なった造血細胞におけるToll様受容体(TLRs)発現の有無を検討した。特にRNAやDNAをリガンドとする細胞内TLR3.TLR7/8, TLR9については、自己免疫疾患と深く関わる分子であると推定されているため、特に重点的に検討した。その結果、これらのリガンドのうち、特にTLR9については研究の急速な進展が見られ、興味深い結果が得られつつある。現在、疾患との関わりも念頭において解析を進めている。本年の日本血液学会および米国血液学会で発表を予定している。また、TLRリガンドが造血に及ぼす影響をin vivoで検証するために野生型マウス(C57BL/6)にTLRリガンドを投与し、骨髄、脾臓、末梢血における変化を検討してきた。既に野生型マウスの実験系が確立した。当初の予想どおり、本実験系ではTLRリガンドを投与された野生型マウスは血球貪食症候群を発症する。現在、その詳細な解析を進めているが、本年度は研究をさらに発展させ、遺伝子改変マウスおよびヒトのin vitroで得られた結果と対比していく。これらの研究によって、血球貪食症候群における樹状細胞の役割はもとより血球貪食症候群の有効な治療法が確立されると考える。本研究の成果は今年度もしくは来年度の米国血液学会で発表する予定である。
