| Research Abstract |
先天性好中球減少症患者の血液細胞を時系列で得て、G-CSF受容体の発現の解析を行い、更にゲノムDNAとRNAを分離、G-CSF受容体細胞膜貫通部分から内側の領域をコードする遺伝子の配列を直接決定する。さらに、先天性好中球減少症との関連の深いELA2, HAX1の遺伝子異常の有無についても解析、この解析と併行してG-CSF受容体刺激伝達系に密接な関連が報告されている制御因子、SHP2、Gab1、Gab2、Grb2、SOCS3、STAT3、STAT5、NRAS、KRAS2、またepigenetic関連因子としてCEBPα、HNRNPE2のmRNAをPCR法で増幅、これを核酸フラグメント解析法により遺伝子変異スクリーニングをし、異常が認められた例について、その異常の内容、出現時期と臨床病態、更にゲノムレベルでの存在の有無を確認、結果と病態との関連を検討する.epigenetic関連因子として、G-CSF受容体の発現を制御する転写因子としてのCEBPα、HNRNPE2遺伝子のプロモーターのDNAメチル化についても、Methylation-specific PCR 法やBisulfitesequencing 法で解析し、病態によって変化するかどうかを解析する。
