Research Abstract

平成21年度は以下の研究成果を挙げた
1)生理的条件下でのGSK3β+/-マウスの骨軟骨組織の解析
ノックアウトマウス由来の骨芽細胞を単離し、同胞野生型由来の骨芽細胞との増殖・分化・生存能についての解析を行った結果、骨芽細胞の増殖・生存については明らかな差を認めなかったが、骨芽細胞の分化においてはALP活性やアリザリンレッド染色の染色性の亢進およびリアルタイムPCR上、I型コラーゲン、オステオカルシンの発現上昇を認めた。さらに、骨芽細胞と造血系細胞の共存培養の結果、破骨細胞形成支持能については、ノックアウトマウス由来の骨芽細胞を用いたもので亢進が認められた。以上のことから、GSK3β+/-マウスにおける骨量の増加はGSK3βの骨芽細胞におけるcell-aoutonomousな作用が関与していると考えられた。
2)病的条件下でのGSK3β+/-マウスの骨軟骨組織の解析
変形性膝関節症モデルをマウスに対し作成し、その進行の程度を評価した結果、GSK3β+/-マウスと同胞野生型マウスの間では明らかな変形性膝関節症進行度の相違を認めなかった。
3)骨芽細胞、軟骨細胞におけるGSK3βの上流および下流のシグナル分子の同定
GSK3βの下流分子として過去に報告のある各種転写因子群の中で、骨軟骨形成に関与が示唆されている分子について検討を行った。具体的にはRunx2について主に解析を行い、GSK3βによりRunx2の発現、細胞内局在に変化はないものの、Runx2の転写活性が抑制されることを明らかとした。
