| Research Abstract |
これまでの自家骨髄幹細胞移植による膀胱再生研究では実験動物としてマウスを使用しており、ICR雄マウスの骨髄幹細胞を採取して、BALB/Cnu/nu雌nudeマウスに移植するという同種移植による実験手法であった。そのため、本年度は昨年度より引き続き、マウスよりも大型のラビットを使用して、同一個体での移植を試みた。
ラビットの大腿骨骨髄より注射針で骨髄由来幹細胞を採取し15% fetal bovine血清および抗生剤附加のDulberco Modified Eagle Mediumの中に入れる。細胞を遠心分離した後に、Type I collagenでコートした培養皿で7日間培養し増殖することが可能であった。
本年度は細胞のマーカーとして、GFP(Green fluorescence protein)発現遺伝子よりも蛍光発現性の長いQtracker labeling kitsを使用して、マーキングした。本年度は、膀胱頚部の再生に注目し、膀胱頚部を粘膜面より-80℃のアイスバーで障害与え、その障害部位へ、30G注射針を使用して培養した細胞1×10^5個/50・1を移植した。
移植後3日目、14日目に膀胱を取り出し、Monoclonal anti-GFP抗体、平滑筋特異抗体を使用して免疫二重染色を行い、レーザー蛍光顕微鏡で移植した細胞の分化、つまり、筋層をはじめとする膀胱頚部の再生を確認した。14日目の膀胱においては、Acta 2 primerを使用してalpha smooth muscle actinを、Myhll primerを使用してsmooth muscle myosin heavy chainを、Real time RT-PCR法にて定量測定し骨髄幹細胞の膀胱平滑筋への分化についての評価を定量的に行うことも可能であった。
