R-spondin1の腸管保護による造血幹細胞移植の成績向上の試み

Research Abstract

R-spondinl(R-Spol)の作用機序について検討を行った。R-Spol 200μgをマウスに3日間投与し、小腸を採取し、定量的PCR法、免疫組織染色を行った。R-Spol投与群では、Wntシグナル標的遺伝子でありるaxin2, lgr5, ascl2の発現亢進がみられた。Lgr5は最近、腸上皮幹細胞であることが報告されており、R-Spolによる腸幹細胞増殖作用を確認するために同じく腸幹細胞マーカーとして同定されたolfm4の染色を行ったところ腸クリプトにolfm4陽性細胞の増加を認めた。これらの結果からR-Spolは直接、腸幹細胞の増殖を刺激することが明らかになった。次に、放射線腸炎モデルにおいて、R-Spolが腸幹細胞の増殖を刺激するかどうかを検討した。マウスに致死量の15Gyの全身放射線照射を行い、その前後3日間、同量のR-Spolを投与した.小腸を採取し、同様な検討を行ったところ,放射線照射によって減少したolfm4陽性腸幹細胞がR-Spol投与群では大幅に増加していることが確認された。以上の結果から,R-Spolは直接的に、腸幹細胞の増殖を刺激することによって放射線腸炎に対する腸管保護作用を有することが明らかとなった.
