新奇テルペン合成酵素の構造と機能に関する研究

Research Abstract

植物が産生するテルペンは、植物に耐熱性、光防護作用、抗菌・抗酸化作用をもたらし、また他の植物、益虫、害虫とのコミュニケーションに利用されている。また食品の質を決める重要な因子でもある。本年度は、その基質特異性の広さから、テルペンアルコールを含む多様なアルコールの酸化する事ができる5つのシンナミルアルコールデヒドロゲナーゼ(CAD)5種について特性解析及び結晶構造解析を行った。5つのCADは一次配列からClassI(主にリグニン合成に関与)とClassII(機能があまり特定されていない)に分類され、そのうちClassIIのCADはゲラニオールに対して高い活性を持っていた。ClassIのCADはシンナミルアルコールなどの基質に対し高い活性を持っていたが、ゲラニオールに対する活性は低かった。ClassIの活性部位の一部をClassII型にすることで、ClassIIよりゲラニオールに対して高い活性を持つ変異ClassI CADの創製に成功した。また、CADファミリーではじめて、基質、補酵素(NADH)との三者複合体のX線結晶構造解析に成功した。基質は、直接活性中心の亜鉛に配位し、補酵素からプロトンを授受するのに都合の良い位置に結合していた。
