胃食道逆流症モデル動物における食道運動の解析

Research Abstract

現在問題となっている食道疾患のひとつが、胃食道逆流症(Gastroesophageal Reflux Disease ; GERD)である。この発症には、胃内容物の逆流が大きく関与する。嘔吐現象と胃内容物の逆流には関係があることは容易に予想できる。一方、現在汎用されているラットやマウスなどの実験動物は、ヒトと異なり、嘔吐しないという特徴がある。そこで、本研究では、生理的に嘔吐する能力を持つ実験動物スンクスを新たな食道疾患モデル動物として応用可能かどうかを検討した。本年度は、昨年度確立したin vitro (ex vivo)実験系を用いて、スンクスの食道運動に関わる神経成分や筋細胞の特性を調べた。
1. スンクスの道運動を支配する神経系
スンクスから食道を摘出し、食道運動を記録した。迷走神経を電気刺激したところ、食道標本の収縮運動が惹起された。この反応は、神経筋接合部の遮断薬で阻害された。この結果は、スンクス食道筋は、迷走運動神経によって支配されていることを示唆している。
2. スンクスの食道筋細胞の特性
スンクスの食道筋は、形態学的に横紋筋であることが明らかとなった。この特徴は、ラットと類似していた。しかしながら、筋細胞上の受容体の薬理学的特徴がラットとは異なっていた。スンクス食道筋細胞の種特異性が示唆された。
