神経因性疼痛における新規細胞間接着因子CHL1の役割

Research Abstract

本研究の目的は神経因性疼痛モデル動物の痛覚伝達系における接着因子Close homologue of L1(CHL1)分子の挙動と疼痛との関連の解明であり、この特色は神経周囲の接着環境の変化に着目して、神経因性疼痛の形成原因を考えている点である。
平成20年度の実験は、主にCHL1に関して発現動態め変化を詳細に検討していくことに主眼を置いた。末梢神経傷害がCHL1分子の挙動にどの程度の変化をもたらすのかを統計的に示すことができるような実験を行った。以下に具体的な実験について述べる。
1. 末梢神経損傷に伴うCHL1の発現変化の詳細な検討
ラットの坐骨神経の損傷モデルにおいて、後根神経節の小型損傷ニューロンおよび脊髄後角においてCHL1陽性の経時的な増加を確認した。
2. CHL1のタンパクの局在を明らかにした。
損傷ニューロンのマーカー(ATF3)や有髄のニューロンのマーカーであるNF200とCHL1の重染色を行った結果、CHL1の発現変化は損傷した無髄線維であることが解った。脊髄においては上記の終末に集積を確認した。
生化学的にはCHL1の切断などの変化は確認できず、CHL1全長蛋白の増加を確認したのみである。
