Research Abstract

2001年に我々が報告したIL-17Fは喘息との関与が示唆されているがその詳細な役割は不詳である。一方、吸入ステロイド薬で喘息の長期管理は容易となってきたが、重症喘息は抗喘息薬の効果は決して高くなく、今後の喘息治療の大きな課題である。その原因の一つとして気道リモデリングが挙げられる。IL-11とIGF-Iは気道リモデリングの形成に深く関与する。重症喘息の病態、特に気道リモデリングの形成メカニズムを解明するためにIL-17Fによる気道上皮細胞からのIL-11とIGF-Iの発現について検討した。その結果、IL-17FはIL-11とIGF-Iを有意に産生し、その発現メカニズムはRaf1-MEK1/2-ERK1/2を上流としたp90RSK-CREBおよびMSK1-CREBの2つの下流シグナル伝達経路が重要であることが明らかになった。IL-17Fやそのシグナル伝達経路が喘息、特に重症喘息の新たな治療標的になる可能性が示唆された。
