Outline of Annual Research Achievements

複雑骨格天然物の新奇な生合成マシナリーの詳細とその立体構造基盤の解明と創薬を目的とした。本研究では、人為的な機能制御と分子多様性創出の格好のモデルともいえる、強力なタンパクリン酸化酵素活性化作用を示すテレオシジンなど、テルペンインドール・アルカロイドとその関連化合物をとりあげた。有機化学を基盤として、その生合成マシナリーの詳細を酵素・遺伝子レベルで明らかにするとともに、酵素反応機構とその立体構造基盤を解明した。また、酵素タンパクの立体構造情報に基づく合理的な触媒機能の改変により、さらなる分子多様性と新規骨格の創出をめざした。本研究により合成生物学と創薬研究の革新的ツールとなる超天然型新規生体触媒と創薬シード化合物の創製を実現をめざした。
2022年度は、カビ由来テルペン合成酵素の機能解析を行い、スクアレンに由来せずに、C5 イソプレン単位ジメチルアリル二リン酸(DMAPP)とイソペンテニル二リン酸(IPP)を基質として、C30トリテルペンの骨格を一挙に構築する、画期的な新奇生合成酵素を世界に先駆けて発見し、論文発表した。加えて、アジリンなどの国家ウを構築する非ヘム鉄2-OG依存性多機能型酸化酵素や関連酵素を派遣し、その立体構造解析と合理的な酵素触媒機能の拡張などに成功した。
