Outline of Annual Research Achievements

本研究では、植物一次細胞壁の主要非セルロース性細胞壁多糖であるペクチンに焦点をあて、「道管細胞分化におけるペクチンダイナミクスをペクチン修飾鍵酵素の観点から明らかにし、道管細胞の通水性制御における非セルロース性細胞壁多糖ダイナミクスの役割を解明する」ことを目的としている。2021年度には、以下の成果を得ることができた。
1. ペクチン分解酵素の酵素特性が細胞分化にどう影響するかを明らかにするため、道管細胞特異的ペクチン分解酵素の欠損変異体に、3種類のペクチン分解酵素を発現させた形質転換シロイヌナズナを新たに作製しラインの整備を行った。作製ラインの予備的解析から、3種類のペクチン分解酵素の性質の違いが細胞分化に影響することを示唆するデータが得られており、今後詳細に調べる予定である。
2. 昨年度同定したペクチン分解阻害タンパク質に着目し、シロイヌナズナから3つの候補遺伝子を同定した。これら3つの遺伝子について、局在を調べるためのレポーター遺伝子を新たに作出し、シロイヌナズナ野生型、シロイヌナズナ道管細胞分化誘導ライン、シロイヌナズナ培養細胞、への導入を行った。また、タバコ葉への一過的発現解析から、これらペクチン分解阻害タンパク質が道管細胞で発現していることを示す結果を得ることに成功した。

各種形質転換体の作出や、新規解析対象遺伝子に関するレポーターの作出など、概ね順調に進んでいる。

Strategy for Future Research Activity

今後は、ペクチン分解酵素の特性評価、形質転換体の形質評価、レポーター観察、などを継続し、予定通りの解析を進める。海外研究協力者との連携を進めながら、道管細胞分化におけるペクチンダイナミクスの解明を進める。
