紫外線照射による制御性T細胞の誘導と解析―皮膚病変のリキッドバイオプシーの開発

Outline of Annual Research Achievements

乾癬、皮膚T細胞リンパ腫(CTCL)の病変部からの皮疹血リキッドバイオプシーを行い症例を集積を行った。皮膚T細胞性リンパ腫19例から、皮疹血リキッドバイオプシーを行い、末梢血とともに解析を行なった。CD4+CD45RO+細胞、CD8+CD45RO+細胞の割合が皮疹血で有意に高く、病変部では、CD8+CD45RO+細胞と、CTCLの皮膚評価スコアであるmSWATが逆相関していた。さらに皮疹血と末梢血の血清を比較すると、CCL17、CCL22など悪性T細胞を誘導するケモカインが皮疹血で高いことがわかった。RNA-seqで発現遺伝子を比較したところ、皮疹血のCD4+CD45RO+細胞ではCTCLの腫瘍細胞に特異的な遺伝子が多く発現していた。TCRレパトア解析にて偏位したT細胞受容体が検出された。皮疹血のCD8+CD45RO+細胞では細胞分裂阻害に関わる遺伝子が多く発現され、末梢血のCD8+CD45RO+細胞では炎症に関わる遺伝子が多かった。CTCLの皮疹血では、末梢血よりも病態に関わる細胞、ケモカインが認められることがわかった。現在、皮疹血からのシングルセルRNAシークエンスを開始し、腫瘍細胞の特異的な遺伝子発現をあきかにすることができた。皮疹血には皮膚疾患特異的な細胞やタンパク質が含まれる可能性があり、他の炎症疾患の診断に役立つ可能性がある。
nTreg(内在性制御性T細胞)の誘導波長解析もついても、モノクロメーターに加え、UV-LEDを用い解析を行い、UVC~UVBが誘導波長であることを見いだした。
