Outline of Annual Research Achievements

長崎原爆被爆者の疫学的検討より原爆放射線によって骨髄異形成症候群（MDS）の発症リスクが上昇することが示されている。原爆放射線がどのようなメカニズムで造血前駆細胞の形質を転換させ、骨髄異形成症候群発症に繋がるのかを検討する目的で、被爆者骨髄異形成症候群のゲノム変異を模したマウスモデルを作成した。これまでの検討で被爆者骨髄異形成症候群では、自然発症例と比較してATM遺伝子の変異頻度が上昇していることが示されている。そこでマウスを用いてatmコンディショナルノックアウトマウスと、自然発症例や被爆者骨髄異形成症候群で共通して変異がみられる遺伝子Bの変異マウスを交配し、薬剤を作用させることで、成体ににおいて造血細胞でのatmおよび遺伝子bとの二重変異マウスを作成した。これまでの報告よりatm欠損マウスは造血器腫瘍としてリンパ系腫瘍を発症することが知られているが、今回作成したモデルマウスの一部は血球減少（貧血）の時期を経た後に造血細胞の異常増殖を示し、骨髄異形成症候群に類似した経過をとって造血器腫瘍を発症した。放射線によって生じたゲノム変異が造血器腫瘍に関連する事を示唆する結果と考えられ、更に詳細な検討が必要である。また、高齢者に一定の頻度でみられるクローン性造血は、骨髄異形成症候群の発症に強く関連すると考えられている。そこで、貧血のみられない被爆者を対象として末梢血におけるクローン性造血について検討を行った。その結果、一般の非被爆者に見られるクローン性造血と異なる特徴を持つことを示唆する結果を得ており、詳細について検討を継続している。
