Outline of Annual Research Achievements

近年、胎児期や乳幼児期の様々な環境要因が成長後の疾患発症リスクにつながるとするDOHaD(Developmental Origins of Health and Disease)概念が提唱されている。しかしながら分子疫学的なエビデンスは未だ十分でない。本研究は大規模出生コホートを用いて、胎児期からの環境要因が学童期の健康リスク特に肥満・やせ、アレルギー、神経発達に及ぼす影響について、遺伝子多型(SNPs)、DNAメチル化および腸内細菌叢の解析からアプローチし、遺伝子ー環境相互作用の観点からDOHaD分子メカニズムを明らかにすることを目的とする。
今年度は母児390組に対して対面調査を行い、質問票調査に加え、血液検体および糞便の回収・保管を行った。SNPsは8歳児および母親に対して解析候補の96SNPsをそれぞれ選定し、解析条件の検討を行い、エコチル解析用SNPチップを構築した。8歳児についてはDNAが抽出できた約1800検体のSNPsデータを取得した。腸内細菌叢(16SrRNA)解析については2021年度までの8歳児の糞便からDNAの抽出を行い、850検体の16SrRNAのデータを取得した。また2021年度分の母児の血清の生化学解析を行いデータを取得した。
また疫学解析から妊娠中の母親の喫煙曝露と3歳時の気管支喘息発症リスクの増加、妊娠中の母親の低タンパク質曝露と3歳時の神経発達遅延リスクの増加をそれぞれ明らかにし、論文発表を行った。今後は取得したゲノム、腸内細菌叢のデータを用いて分子疫学的解析を行う予定である。

コロナ禍やウクライナ情勢の影響で試薬の入手に時間がかかったことから、当初の計画からはやや遅れている。

Strategy for Future Research Activity

母親の糞便からDNAを抽出し、16SrRNAのデータを取得する。
母親のSNPsデータを取得する。
2022年度分の母児の血清生化学解析を行う。
得られたSNPsおよび16SrRNAデータを用いて、胎児期・乳幼児期の環境要因と学童期の健康リスクへの影響を解析する。
