Outline of Final Research Achievements |
We demonstrated that cells have mechanisms to tolerate for illudin S-induced DNA lesions. Importantly, the tolerance required PCNA ubiquitination but independently of DNA polymerase eta. We found that DNA polymerase kappa and RFWD3 play crucial roles independently to each other, but both dependently on PCNA ubiquitination. We published "RFWD3 and translesion DNA polymerases contribute to PCNA modification-dependent DNA damage tolerance".

Academic Significance and Societal Importance of the Research Achievements |
DNA 損傷トレランスはゲノム安定性の維持に重要なメカニズムである。一方で、DNA損傷による DNA 複製阻害を引き起こすことで細胞死を誘導する、シスプラチンのような抗がん剤に対しては、耐性に寄与するという側面があり、がん細胞の増殖に寄与する。従って、RFWD3 の DNA 損傷トレランスにおける詳細な機能を解析するとともに損傷乗り越えDNA合成との連携機構などを解明することにより、ゲノム安定性を維持するしくみだけなく、がん細胞の抗がん剤耐性のメカニズムの解明及びその克服法や、新たな抗がん戦略の構築につながることが期待される。
