シクロメタレート型イリジウム（Ir(III)）錯体は、tpy (= 2-(4’ -tolylpyridine)) などのリガンドがCとNを介してIrに配位した錯体である。Ir錯体は一般的に水中で非常に安定であり、三重項励起状態を経由した強いりん光を発する。申請者らは、Ir錯体にぺプチドを導入したIr錯体―ペプチドハイブリッド（Ir complex-peptide hybrid, IPH）を設計、合成し、 IPHが、がん細胞の細胞死を誘導して、死細胞中で強く発光することを見出した。そして、細胞の形態変化観察などにより、その細胞死は、代表的なプログラム細胞死（PCD）であるapoptosisではなく、小胞体やミトコンドリアの液胞化を伴うparaptosisであることを見出した。paraptosisは近年発見されたPCDの一つであり、天然由来のcelastrolなどによるparaptosisの誘導が報告されている。そこで本研究は、申請者らのIPHとcelastrolなどを用いて、paraptosis機構の解析を行った。(i) IPHとcelastrolによって誘導された細胞の透過型電子顕微鏡(TEM)画像が酷似していること、(ii) IPHはリソソームや小胞体の液胞化を誘導するが、celastrolはリソソームの液胞化を誘導する、(iv) IPHはミトコンドリアと小胞体の膜融合を誘導するが、celastrolは誘導しない、(v) IPHはミトコンドリア内Ca2+濃度上昇を誘導し、celastrolはミトコンドリアと細胞質内のCa2+濃度を上昇させる、などの違いが観察された。両者のparaptosis誘導機構が異なることから、celastrolによって誘導されるPCDをparaptosis I、IPHによって誘導されるPCDをparaptosis IIと分類した。
