Outline of Final Research Achievements |
In this study, we generated the intestinal organoids from human iPS cells and were able to develop the method in which the cells were able to be cultured with maintaining crypt-villus structures by the two-dimensional culture of the intestinal organoids. We could obtain preliminary findings to be suggested that the intestinal mucosal damage could be able to be evaluated by using the two-dimensional cultured cells. Additionally, we were able to develop the differentiation method into the endodermal cells from human iPS cells efficiently and the culture method in which the two-dimensional intestinal organoids can be cultured for a relatively long term with keeping crypt-villus structures.

Academic Significance and Societal Importance of the Research Achievements |
本研究ではヒトiPS細胞から腸管上皮細胞および腸管オルガノイドを作製し、疾患や医薬品によるの消化管傷害を評価可能なin vitroモデル系を構築することを目的として研究を進めた。その結果、ヒトiPS細胞由来腸管細胞が消化管の粘膜障害を評価するためのin vitroモデル系としての応用の可能性を示唆することができた。したがって、本研究では創薬研究等での利用に向けて有用な知見を得ることができたと考えられた。
