Outline of Final Research Achievements

Preparation of a pyrG gene disrupted strain from Paraboeremia sp. Lsl3, a Lycopodiaceae plant-drived endophytic fungus producing Lycopodium alkaloids, was tried, and preparation of ΔpyrG strain, ΔligD/ΔpyrG strain, and ΔligD/ΔargB strain of Aspergillus oryzae as heterologous expression hosts has been succeeded. Furthermore, a Lycopodium alkaloid-like novel skeleton compound has been discovered from a transformant along with pelletierine.

Academic Significance and Societal Importance of the Research Achievements

Lycopodiumアルカロイドは、アルツハイマー病改善薬候補であるhuperzine Aに代表される構造多様性に富んだ化合物群であり、その骨格構築に関するメカニズムは多くの研究者の興味を集めている。本研究では、生合成遺伝子の異種発現において、Lycopodiumアルカロイド様の新奇化合物を発見した。今回得られた知見は、植物由来の複雑骨格天然物を構築する同様のメカニズムを微生物が保有することをこれまで以上に強く示した。
