Outline of Annual Research Achievements

昨年度までに、アルツハイマー病モデルマウスとして、APPV717I::TAUP301Lマウスを作出し、さらにNrf2活性化アルツハイマー病モデルマウスとしてAPPV717I::TAUP301L::Keap1FA/FAおよびAppNLGF::Keap1FA/FAマウスを作出し、グルタチオンの脳内分布、およびグルタチオン代謝に関連するアミノ酸代謝について解析を行い、さらにアルツハイマー病モデルマウスにおけるNrf2活性化剤CDDO-2P-Imの脳内移行について確認を行った。最終年度にあたる今年度は、アルツハイマー病モデルマウスにCDDO-2P-Imを投与し、アルツハイマー病に関連する影響を検討した。
APPV717I::TAUP301LマウスにCDDO-2P-Imを投与したところ、脳内のグルタチオンレベルが上昇した。さらに、APPV717I::TAUP301Lマウスでは、脳にアミロイドβ(1-42)が蓄積し、野生型マウスと比較して高いレベルであったが、同マウスにCDDO-2P-Imを投与したところ、アミロイドβ(1-42)の脳内レベルの強力な低下を認めた。同様にAppNLGF::Keap1FA/FAマウスを解析したところ、野生型マウスと比較してAppNLGFマウスで高いレベルの脳アミロイドβ(1-42)を認め、AppNLGF::Keap1FA/FAマウスではAppNLGFマウスと同レベルであった。以上より、APPV717I::TAUP301Lマウスにおける薬剤によるNrf2活性化では、グルタチオンが増加することが確認された。さらに、APPV717I::TAUP301Lマウスにおける薬剤によるNrf2活性化では、アルツハイマー病の重要な原因となるアミロイドβ(1-42)が低下することを同定した。
