Outline of Annual Research Achievements |
Neurodome culture was performed routinely in the last year, and both application of ApoE4 as well as application of tau pre-formed-fibrils showed increase in amyloid plaque formation and hyper-phosphorylated tau. This pathology manifests significantly more in the Neurodome cultures than in conventional cultures. In addition, in collaboration with another researched from our unit (Zacharie Taoufiq, equal contribution), we filed a patent application on 2021/11/30 as follows, Japan Patent Application#2021-194719, Proteimics-Based Receptor-Ligand Matching For Optimizing iPS Cell Reprogramming.

2: Research has progressed on the whole more than it was originally planned.
Reason
Currently, I am continuing to gather data from the neurodome cultures and quantifying the amount of amyloid plaques and phosphorylated tau, and comparing to conventional cultures. I am also performing the same experiments on an additional iPS cell line. I am in the process to obtain a third cell line from a cell bank. , and perform the same experiments. Having the same experiments and results on more than one cells lines would strenghten the utility of the Neurodome culture, and show that this method in applicable to a broad range of cell lines.

Strategy for Future Research Activity |
The main goal is to prepare and submit a manuscript in an high-impact international journal. As soon as I can show that neurodome cultures can be made from 3 different and independent iPS cell lines, and that Neurodome cultures consistenly can show Alzheimer disease pathology (amyloid plaques and tau hyper-phosporylation), I will begin to prepare a manuscript and submit to a good journal.

Causes of Carryover |
International travel was stopped in the last two years. Also, some research products took longer to arrive to Japan, so purchases of products were delayed by a few months and thus products planned for last fiscal year, will be bought this fiscal year.
