Outline of Annual Research Achievements |
We previously reported that major white matter tracts were impaired in the Sulf1/2 double-knockout (DKO) mice. Then, we investigate the topology of the brain microvasculature in the cortex of mice lacking endosulfatases. We performed a vascular casting with a newly developed method called SeeNet. We acquired 3D confocal images dataset in the motor, somatosensory and visual cortex. Using the filament tracer tool in Imaris software, we reconstructed the whole vascular network. Then, data were then exported and analyzed with Python and R software. We investigated spatial vessel density, vessel diameter, branchpoint density, and tortuosity. We identified fine variation in microvascular density and topology based on cortical location and depth. The visual cortex consistently showed lower vascular density, especially in the deeper layer. In addition, we found that vascular topology in the Sulf1/2 DKO was different than in the wild type cortex. While vascular density was consistent, we found a significantly higher branchpoint density in the Sulf1/2 DKO throughout all cortical region and depth.
