Outline of Final Research Achievements

Colitis-associated cancer (CAC) has clinicopathologically and genetically different characteristics from sporadic colorectal cancer. Next-generation sequencing-based gene panel testing revealed that CAC has different genetic profile from sporadic colorectal cancer. In addition, it revealed characteristic mutational signatures which could be useful for treatment in CAC. It was suggested that these genetic features of CAC could be applied to the diagnosis and treatment of CAC.

Academic Significance and Societal Importance of the Research Achievements

罹病期間の長い炎症性腸疾患では、一定の頻度でCACが発生する。CACの診断および治療に関する臨床的な課題として、診断が困難であること、薬物療法が確立していないことが挙げられる。本研究の学術的意義は、次世代シークエンサーによる網羅的な遺伝子変異解析を用いて、CACに特徴的な遺伝子変異プロファイルを明らかとしたことである。さらに、バイオインフォマティクスによる変異シグネチャー解析によって、治療に応用可能な変異シグネチャーを検出したことである。本研究の社会的意義は、CACの診断および薬物療法の体系を確立するための基盤となるCACの遺伝子学的特徴を明らかとしたことである。
