Outline of Annual Research Achievements

これまでの研究で、ヒトとウシの歯のエナメル質と象牙質の結晶性ナノ構造に違いがあるものの、フッ化物への反応性に大きな差異があることが判明した。特にエナメル質は既に低い反応性を示すため、2%の高濃度フッ化物を短時間塗布しても歯質改善効果は少ないと推測された。フッ化物の歯質強化および脱灰抑制の作用を調べるために、19F核磁気共鳴(NMR)および19F固体(SS)魔法角回転(MAS)NMRを用いた研究を行った。
使用した高濃度フッ化物は、i) 2%中性フッ化ナトリウム(N-NaF)、ii) 2%酸性フッ化ナトリウム(A-NaF)、iii) 38%フッ化ジアンミン銀(SDF)の3種類である。ウシエナメル質粉末を各フッ化物製剤で処理後、脱水して19F SS-MAS NMR測定を行った。酸性、中性、アルカリ性条件下でのCaF2のフッ化物徐放性を19F NMRで評価したところ、すべての製剤で水性F-のピークが確認された。SS-MAS NMRとスペクトルデコンボリューションにより、すべての処理エナメルからフルオロアパタイトとCaF2の2つの主要なピークが検出された。N-NaFとA-NaFを比較すると、A-NaFの方がより強いシグナルを示した。一方、SDFではアルカリ性の影響でフッ化物生成が少なかった。いずれの処理でもフルオロアパタイトとCaF2が生成され、特にA-NaFは酸性であるためエナメル質表面を溶解し副生成物を生成しやすいことが示された。CaF2は様々な条件下で微量のフッ化物を長期間放出する可能性があるが、そのreservoirとしての役割には疑問が残った。本研究の結果は、2024年3月のIADR国際大会で口頭発表(19F-NMR Study of Fluoride Reaction of Various Formulations on Enamel)にて報告された。
