Outline of Annual Research Achievements

異所性動物実験(生物検定): Wistar 系ラット頭頂骨を切除して試料骨片(5mmx5mm)を5群調整した.
1. 穿孔骨 (バー φ0.6mm), 2. 超音波/ 生理食塩水処理骨(120W, 38kHz, 20min, 25°C),3. 強酸性電解水(pH2.7)単独処理骨, 4. 超音波/強酸性電解水(pH2.7)処理骨, 5. 未処理骨(新鮮骨)

生物検定:各種骨片を同種Wistar 系ラット(4週齢,雄性)背部皮下組織内に移植した.
評価観察方法:移植後7,14,21,28日目に周囲軟組織とともに摘出した.
固 定方法:20%中性ホルマリン溶液に浸漬後にマイクロウェーブを加えて固定液の組織内浸透性を高めた.

計画に基づき,異所性(背部皮下組織内)動物実験を終了できた.

Strategy for Future Research Activity

20%中性ホルマリンに浸漬固定中の試料を脱灰して標本を作成する.
固定方法:マイクロウェーブを加えて固定液の組織内浸透性を高める.
脱灰方法: 10% 蟻酸溶液で14日間脱灰後,連続切片(4μm)を 作製する.
染色方法:HE染色・II 型コラーゲン染色で骨・軟骨誘導を観察し,組織形態計測で移植骨と誘導組織の割合を経時的に計測する.
