Outline of Final Research Achievements

In this study, we developed a method to evaluate the TLR9-independent innate immune activation of antisense oligonucleotides. We showed the need for evaluation methods using human cells, and generated reporter cell lines to evaluate innate immune activation.

Academic Significance and Societal Importance of the Research Achievements

本研究は動物試験あるいは既存のin vitro試験法（ヒトTLR9発現細胞）では評価できない自然免疫活性化経路を初めて提唱する重要な成果であると考えている。核酸医薬品を含め、核酸を用いた医療技術（ｍRNA関連医薬、遺伝子治療薬等）の自然免疫系の評価については、種差の観点から動物試験での予測が難しいとされるが、これに対して、本研究はヒト培養細胞を用いた新しい評価技術を創出するものであり、社会ニーズへの対応、医療分野での貢献という点でも極めて有用な成果であると考えている。
