Outline of Final Research Achievements |
There have been several reports about human papillomavirus (HPV)-derived antisense RNAs expressed in HPV-related cancers but their detailed structures and functions have been unknown. Therefore, this study aims to identify the structures and functions of HPV-derived antisense RNAs. PCR cloning and subcellular fractionation experiments demonstrated that HPV antisense RNAs expressed in certain HPV-related cancer cells are a type of nuclear long noncoding RNAs (termed as HPV-AS lncRNA). Knockdown analysis revealed that HPV-AS lncRNA supports cancer cell survival in nutrient/growth factor-scarce environments and resistance to oxidative stress, suggesting that it possesses functions required for metastasis.

Academic Significance and Societal Importance of the Research Achievements |
長鎖非コードRNAはがんの分子マーカーの有力な候補として考えられているが、HPV関連がんにおける基礎生物学的知見は不足している。本研究はHPV由来の新たな長鎖非コードRNAを同定し、その機能の一端を解明したことから、HPV関連がんにおける長鎖非コードRNAの研究に新たな知見を加えるものとなっている。本研究で同定した長鎖非コードRNAはがん転移の細胞機序に関わることから、がん転移の診断マーカーの検討や転移を抑制するための薬剤の開発などを行うために必要な基礎学術的知見の集積に貢献すると考えている。
