胎児期化学物質曝露による真のエピゲノム影響の評価

Outline of Annual Research Achievements

胎児期の環境要因が将来の健康や病気の発症リスクに影響する”というDOHaD(Developmental Origins of Health and Disease)の概念が定着している。その分子メカニズムとしてエピジェネティクス特に、DNAのメチル化変化に焦点があてられている。一方で、DNAの化学修飾にはヒドロキシメチル化も知られ、遺伝子発現促進に関与していることも報告されている。したがって真のエピゲノム変化を明らかにするためにはDNAのメチル化とヒドロキシメチル化の両面から解析する必要がある。
本研究は、化学物質曝露の中でもエピゲノムへの影響が大きいとされる喫煙曝露により影響を受ける真のDNA修飾変化をもたらす遺伝子領域を明らかにすることを目的とした。
出生コホートにおいて、妊娠中の喫煙曝露と網羅的臍帯血DNAのメチル化を解析から有意にメチル化変化する領域を同定し、変化の大きかったAHRRおよびGFI1遺伝子について次世代シークエンサーでメチル化及びヒドロキシメチル化解析を行った。その結果、喫煙暴露とメチル化は有意な変化が確認できたが、ヒドロキシメチル化との関連は見られなかった。さらにヒトiPS細胞を用いて、タバコ煙曝露によるグローバルなメチル化およびヒドロキシメチル化変化の影響を解析した。その結果、ある曝露濃度においてメチル化とヒドロキシメチル化の割合が逆相関することがわかった。したがって喫煙曝露によりヒドロキシメチル化が変化する遺伝子領域が存在することが示唆された。今後具体的な遺伝子領域の同定、機能への影響を明らかにする必要がある。
