Outline of Final Research Achievements

Genes in the nucleocidin biosynthetic gene cluster were individually deleted by genetic manipulation. Enzymes responsible for the initial steps during the biosynthesis were characterized by in vitro assays. Based on the results, a possible biosynthetic pathway has been proposed.

Academic Significance and Societal Importance of the Research Achievements

ヌクレオシド骨格は抗ウイルス薬や抗生物質などの医薬品に頻繁にみられる化学構造であり、生物活性の発現に重要な役割を果たしている。 本研究により得られた生合成情報をもとにヘテロ原子を含む希少ヌクレオチド天然物の新規生合成経路や新規酵素の発見が期待できる。また、同定した生合成酵素あるいは適切なアミノ酸置換を施した酵素変異体を用いることにより新規生物活性ヌクレオシド化合物の創出も期待できる。
