As an intrinsically disordered region (IDR), the overall structure of EML4-ALK remains unclear. This study imaged the dynamic structures of variant EML4-ALK fusions using a high-speed atomic force microscope. Our data suggested that EML4-ALK displayed monomer, dimer, and trimer in solution conditions. Variant EML4-ALK displayed different monomeric profiles which guided structural differences in trimers, especially in the trimers of variant-3. Thus, this study might reveal a new mechanism of the specific chemoresistance based on the structural dynamics.
